چکیده

The homeobox gene superfamily is highly conserved during evolution. These genes act as transcription factors during several important developmental processes through their 60 amino acid homeodomains that recognize and bind to the regulatory region of their target genes. We isolated a novel homeobox gene designated Tex1 from a mouse testis cDNA library by degenerate oligonucleotide polymerase chain reaction (PCR) screening. Three extra amino acid residues, AYP, are inserted between the first and second helices of the homeodomain, which makes Tex1 a TGIF subclass homeobox gene. Tex1 was expressed specifically in the testis as seen by both Northern blot and RT-PCR analysis. Tex1 expression is initiated at around day 20 of testes. Furthermore, the results of in situ hybridization analysis reveal that the expression of Tex1 in testis is restricted in the germ cells at spermatid stage. Based on these data, it is suggested that Tex1, a novel testis homeobox gene, may play a crucial role during spermatogenesis.

Identification of Spata-19 New Variant with Expression beyond Meiotic Phase of Mouse Testis Development

Background: The study of specific genes expressed in the testis is important to understanding testis development and function. Spermatogenesis is an attractive model for the study of gene expression during germ cell differentiation. Spermatogenesis associated-19 (Spata-19) is a recently-identified important spermatogenesis-related gene specifically expressed in testis. Its protein product is in...

Spx1, a novel X-linked homeobox gene expressed during spermatogenesis

Spx1, a novel mouse homeobox gene, encodes a homeodomain characteristic of the paired-like class of homeobox genes and has been mapped to the distal end of the X chromosome. Northern blot hybridization of adult tissues detected high levels of a single Spx1 transcript in the testis. Further analysis by in situ hybridization revealed predominant Spx1 expression within the spermatogonia/preleptote...

Study of Tnp1, Tekt1, and Plzf Genes Expression During an in vitro Three-Dimensional Neonatal Male Mice Testis Culture

Background: In vitro spermatogenesis has a long research history beginning in the early 20th century. This organ culture method was therefore abandoned, and alternative cell culture methods were chosen by many researchers. Here, whether Tnp1, Tekt1, and Plzf, which play a crucial role in spermatogenesis, can be expressed during testis organ culture was assessed. Methods: Testes of 10 mouse pups...

Requirement for two nearly identical TGIF-related homeobox genes in Drosophila spermatogenesis.

The genetic analysis of spermatogenesis in Drosophila melanogaster has led to the identification of several genes that control the onset of meiosis, spermatid differentiation, or both. We described two tightly linked and nearly identical homeobox genes of the TGIF (TG-interacting factor) subclass called vismay and achintya that are essential for spermatogenesis in Drosophila. In flies deficient...

P-230: Analysis of TEX15 Expression in Testis Tissues of Severe Oligozoospermic and Non-Obstructive Azoospermic Men Referred to Royan Institute

Background: TEX15 is a novel protein that is required for chromosomal synapsis and meiotic recombination. Human TEX15 is located on chromosome 8(8p12 region) and expressed in testis and ovary, as is its mouse ortholog. Loss of TEX15 function in mice causes early meiotic arrest in males but not in females. Specifically, TEX15 deficient spermatocytes exhibit a failure in chromosomal synapsis. In ...
